Purpose:
|
To determine the possible interactional association of high sensitivity CRP levels and CCL2–2518 gene polymorphism with wet AMD and evaluation of their possible interactive role in AMD susceptibility.

Methods:
|
137 patients with wet type AMD and 161 healthy controls were recruited and examined. Serum samples were assayed for high sensitivity (hs) CRP which was measured with a high-sensitivity assay. Peripheral blood sample was obtained from all subjects for DNA extraction. Polymorphism in CCL2–2518 (rs 1024611) was genotyped using polymerase chain reaction and restriction fragment length polymorphism .

Results:
|
The genotype and allele frequencies for CCL2 were not found to be significantly different between AMD and normal controls. Regarding Non-GG and Low CRP (CRP≤3mg/dl) as reference group, non GG-high CRP revealed risk ratio of OR=0.53, GG-low CRP combination showed risk ratio of OR=0.63 and GG-high CRP combination showed OR= 0.75 for wet AMD.

Conclusion:
|
There was no difference in hs CRP and CCL2 (-2518 A/G) genotypes between study groups. Our results showed no synergistic or additive activity between these two factors for wet AMD susceptibility.
